Eritrea is scheduled to compete at the 2016 Summer Olympics in Rio de Janeiro, Brazil, from 5 to 21 August 2016. This will be the nation's fifth consecutive appearance at the Olympics.

- Men's 5000 m – 2 quota places (Abrar Osman, Aron Kifle)
- Men's 10000 m – 2 quota place (Abrar Osman, Nguse Amlosom)
- Men's marathon – 3 quota places
